Semaglutide is a synthetic agonist of the GLP-1 receptor (glucagon-like peptide-1), structurally derived from native human GLP-1 with 94% sequence homology. Its key structural modifications include the substitution of alanine at position 8 with 2-aminoisobutyric acid (Aib), as well as C18 acylation linked to lysine via a γGlu-mini-PEG dual-chain spacer.

These modifications give semaglutide marked enzymatic resistance to dipeptidyl peptidases (DPP-4) and high serum albumin affinity — parameters of interest in in vitro pharmacokinetic studies. This compound is used as a reference ligand in GLP-1R radiolabeling and receptor competition protocols.
